Research findings
- Full-length thymosin β4 is characterised as the principal intracellular G-actin sequestering peptide, binding actin monomers at approximately 1:1 and inhibiting polymerisation to F-actin [1, 3]
- Identified as an actin-sequestering peptide, indistinguishable from the previously described Fx peptide [1]
- Found to sequester the majority of G-actin in resting human neutrophils, at cytoplasmic concentrations around 150 µM [2]
- Shown alongside thymosin β10 to bind skeletal muscle actin and inhibit polymerisation, with Kd values of roughly 0.7–1 µM [3]
- All of the above concerns the 43-residue parent protein, not the 7-residue fragment supplied; we are not aware of published work establishing equivalent actin-binding behaviour for the fragment
